Semaglutide Split Dose Calculator

Thinking of taking Semaglutide twice a week? Enter your weekly dose, the days you would inject, and your vial to see the dose and syringe units for each injection, and how the modelled peak and trough compare with one weekly injection of the same total.

Label schedule: The Wegovy label gives the injection once a week. This tool models what splitting the weekly dose would do to drug levels; it does not recommend splitting.

Doses in One Vial 10injections (5 weeks of supply)

By volume this vial holds 10 injections, but FDA generally recommends using multi-dose vials of sterile medications within 28 days of first use: on this schedule that is 8 injections (4 weeks).

Split as 0.5 mg on Monday and Thursday, a 1 mg weekly dose of Semaglutide is 30 units per injection at 1.67 mg/mL. In the model, the steady-state peak is 9% lower and the trough 17% higher than one 1 mg injection a week.

Units assume a 5 mg vial mixed with 3 mL of bacteriostatic water (1.67 mg/mL). Peak-to-trough is the highest level of the week divided by the lowest; 1.00 would be a flat line.

Twice-weekly Semaglutide split: units per injection

Each weekly dose halved, for a 5 mg vial mixed with 3 mL (1.67 mg/mL) on a U-100 syringe. Change the vial above for your own numbers.

Weekly dose Each of 2 injections Units to draw
0.25 mg 0.125 mg 7.5 units (0.075 mL)
0.5 mg 0.25 mg 15 units (0.15 mL)
1 mg 0.5 mg 30 units (0.3 mL)
1.7 mg 0.85 mg 51 units (0.51 mL)
2 mg 1 mg 60 units (0.6 mL)
2.4 mg 1.2 mg 72 units (0.72 mL)
7.2 mg 3.6 mg 216 units (2.16 mL)

Any result over 100 units is more than one U-100 syringe holds. A higher concentration (less water in the vial) gives a smaller volume.

Quick answer: The Wegovy label gives the injection once a week. Split twice a week, a 1 mg weekly dose is 0.5 mg per injection (30 units at 1.67 mg/mL). In the model the weekly total and the total exposure stay the same, the peak is 9% lower and the trough 17% higher than one weekly injection.

Can you take Semaglutide twice a week?

The Wegovy prescribing information (semaglutide) says:

"Administer WEGOVY injection once weekly, on the same day each week, at any time of day, with or without meals."

Source: WEGOVY (semaglutide) Prescribing Information, Novo Nordisk, section 2.1 (DailyMed, effective 2026-06-18).

The label's schedule is one injection a week. It gives no twice-weekly or split schedule, so a split schedule is a departure from it. This page shows what the model says a split would do to drug levels. It is not a recommendation to split; that is a decision for you and your prescriber.

The label also sets a minimum gap between two doses, for when you move your weekly dose to another day:

"If you need to change the day of the week, you may do so as long as your last dose of WEGOVY was given 2 or more days before."

Source: WEGOVY Medication Guide (DailyMed, effective 2026-06-18).

That gap is about moving one weekly dose; the label does not address splitting a dose, so it does not tell you whether any split schedule is acceptable. The label's gap is elapsed time, while this calculator counts whole calendar days, so it marks any two injections 2 days apart or closer.

What splitting changes, and what it does not

Splitting divides the same weekly dose into smaller injections. The weekly total does not change, and in the model neither does the total exposure over the week (the area under the curve): a 1 mg week is a 1 mg week however you divide it. What changes is the shape. With a half-life of about 7 days, a smaller dose every few days keeps the level from climbing as high after each injection or sagging as low before the next one.

In the model, Monday and Thursday puts the steady-state peak 9% lower and the trough 17% higher than one weekly injection, moving the peak-to-trough ratio from 1.61 to 1.25. Daily injections flatten it to 1.02. With evenly spaced injections, each extra injection a week buys less smoothing than the one before; with uneven days it depends on which day you add.

Why the days you pick matter

Only the gaps between injections matter, not the weekday names: Sunday and Wednesday behaves like Monday and Thursday. Uneven gaps let the level sag further over the longest one, so Monday and Thursday (a 3-day and a 4-day gap) gives 1.25, while an even split every 3.5 days, such as Monday morning and Thursday evening, gives 1.19. Two injections close together and a long gap after them, such as Monday and Tuesday, smooth the curve much less. Tap different days in the calculator to see it.

What this tool does not tell you

It does not tell you whether to split your dose. It models population-average drug levels, not your blood levels, and it says nothing about side effects or how well the medication works: a lower modelled peak is not evidence that nausea or weight loss changes. Smaller injections also mean smaller draws, so check every draw against your own vial. The "Doses in One Vial" figure counts by volume only: FDA generally recommends using multi-dose vials of sterile medications within 28 days of first use, and this tool does not tell you how long an opened vial stays usable.

Frequently asked questions

Can you take Semaglutide twice a week?

The Wegovy prescribing information (semaglutide) says: "Administer WEGOVY injection once weekly, on the same day each week, at any time of day, with or without meals." The label's schedule is one injection a week; it gives no twice-weekly or split schedule, and this tool does not recommend one. The label's minimum gap between two doses ("If you need to change the day of the week, you may do so as long as your last dose of WEGOVY was given 2 or more days before.") is for moving the weekly dose to another day; it does not address splitting a dose. In the model, a Monday and Thursday split of the same weekly dose puts the steady-state peak 9% lower and the trough 17% higher than one weekly injection: a peak-to-trough ratio of 1.25 instead of 1.61.

How much Semaglutide is each injection if I split my dose?

Divide the weekly dose by the number of injections. 1 mg a week as two injections is 0.5 mg each; with a 5 mg vial mixed with 3 mL of water (1.67 mg/mL) that is 30 units (0.3 mL) on a U-100 syringe. The weekly total does not change. Enter your own dose, days and vial above.

Does splitting lower the peak?

In the model, yes. Twice weekly (Monday and Thursday) the peak is 9% lower and the trough 17% higher than once weekly; daily injections give a peak 16% lower and a trough 33% higher. The total amount over the week (the area under the curve) is the same for every schedule with the same weekly dose: splitting reshapes the curve, it does not add or remove drug.

Is Monday and Thursday the same as every 3.5 days?

Close, but not quite. Monday and Thursday leaves a 3-day gap and a 4-day gap, and the level sags further over the longer one: a peak-to-trough ratio of 1.25. Injecting every 3.5 days (for example Monday morning and Thursday evening) gives 1.19. Only the gaps matter, so Sunday and Wednesday is the same as Monday and Thursday.

Does splitting reduce nausea or change how well it works?

This tool cannot tell you. It models drug levels, not effects or side effects, and a lower modelled peak is not evidence that nausea or weight loss changes. Talk to a clinician before changing how you take a medication.

How is this calculated?

A one-compartment model with first-order absorption, the same model as the GLP3 Planner half-life calculator, for the injection. For Semaglutide: half-life about 7 days (Wegovy label: approximately 1 week) and time to peak 2 days (Wegovy label: 1 to 3 days after a dose; the model uses the midpoint). The weekly dose is divided evenly over your injection days, every dose's curve is added together until the weekly pattern repeats, and the highest and lowest points of that week are compared with the same total taken once a week. Check: no schedule can swing more than instant absorption allows, 2^(7 / 7) = 2.00 once weekly; the model gives 1.61. The percentages do not depend on your dose or on how much of the dose is absorbed.

Safety: Semaglutide is the active ingredient in FDA-approved GLP-1 medications. This tool is educational only. It models population-average drug levels, not your own, and it does not recommend a dosing schedule or tell you to split a dose. Talk to your prescriber before changing how you take a medication, and double-check every draw against your own vial.

ii About the Model

What drives the curve:

  • Half-life: about 7 days (Wegovy label: approximately 1 week)
  • Time to peak: 2 days (Wegovy label: 1 to 3 days after a dose; the model uses the midpoint)
  • Same weekly total, split evenly
  • Steady state: after weeks on the same schedule
